Apja kívánságára jogot és közgazdaságtant tanult. 1838-ban elhatározta, hogy matematikus lesz. Münsterben fejezte be egyetemi tanulmányait, utána a braunschweigi gimnáziumban tanított matematikát és más tárgyakat 15 évig. A tanítás mellett rendszeresen foglalkozott matematikai kutatással. 1854-ben közölte első cikkét az Abel-féle függvényekről a Crelle's Journal-ban. Eredményeire felfigyelt a königsbergi egyetem, 1854-ben tiszteletbeli doktorrá avatta. Meghívást kapott a berlini műszaki főiskolára, majd néhány évvel később a berlini egyetem professzora lett. Fő céljának az analízis szilárd megalapozását tekintette. Ő adta az első szigorú definíciót a határérték, differenciálhányados, differenciálhatóság és konvergencia fogalmára, a hatványsorok konvergenciájának feltételeire. Ő adta az első példát mindenütt folytonos, de seholsem differenciálható függvényre. Weierstrasst ezért a "modern analízis atyjá"-nak tekintjük. Számos tétel névadója (Weierstrass tétele, Weierstrass approximációs tétele). Életének szinte utolsó pillanatáig folytatta előadói és kutatói munkásságát. Kummerrel és
Kroneckerrel együtt a berlini egyetemet a matematikai élet egyik centrumává tette.
