Az amsterdami egyetemen 1894-ben védte meg doktori disszertációját, melyet Korteweg irányítása mellett készített. Ebben fogalmazta meg a folyadékfelszínen terjedő szolitonokat leíró, ma Korteweg-De Vries egyenletnek nevezett, alapvető fontosságú modellt. Sajnálatos módon az eredmény jelentőségét csak jóval később ismerték fel. De Vries középiskolai tanárként működött nyugdíjbavonulásáig, 1931-ig.
